Biofield Energy Treated Vitamin D3: Direct Effect on Bone Health Using Human Osteoblast-Like Cells

Abstract: Vitamin D and calcium are one of the important constituents of good bone health. Bone disorders might be to the hormonal factors and lack of proper nutrition that leads to gradual bone loss. The current study aimed to explore the potential of Consciousness Energy Healing based vitamin D3 and DMEM medium on various bone health parameters. Vitamin D3 and DMEM medium as test items (TI), were divided into two parts. Each sample received the Consciousness Energy Healing Treatment by Sheila Maureen Benson and those samples were labeled as the Biofield Energy Treated (BT) samples, while the other parts of each sample were denoted as the untreated test items (UT). Bone health biomarkers such as alkaline phosphatase enzyme (ALP) activity, collagen levels and bone mineralization were measured. MTT data showed that cell viability was more than 70% and all the test items were found to be safe at the tested concentrations. ALP level was significantly increased by 135.9% and 134.5% at 1 and 50 µg/mL, respectively in the UT-DMEM+BT-TI, while 84.4%, 93.8%, and 50.0% at 1, 50, and 100 µg/mL, respectively in BT-DMEM+UT-TI group. In addition, BT-DMEM+BT-TI group showed increased ALP by 50.2% and 104.7% at 10 and 50 µg/mL, respectively as compared with the untreated test item and DMEM group. Collagen content was significantly increased by 463.6%, 387.9%, and 239.4% in the UT-DMEM+BT-TI, BT-DMEM+UT-TI, and BT-DMEM+BT-TI group, respectively at 0.1 µg/mL as compared with the untreated group. Moreover, the percent of bone mineralization was significantly increased by 131.7% and 81.7% at 1 and 10µg/mL, respectively in UT-DMEM+BT-TI group, while 83.1% and 53.6% at 10 and 50µg/mL, respectively in BT-DMEM+UT-TI group as compared with the untreated group. In addition, BT-DMEM+BT-TI group showed a significant increased bone mineralization by 152.3% and 108.3% at 50 and 100µg/mL, respectively as compared with the untreated group. Overall, the experimental data suggested that the Biofield Energy Treated vitamin D3 and DMEM would play an important role in the promotion and maintenance of strong and healthy bones, which improve quality of life. Biofield Energy Treatment might also regulates the osteoblast function, improves bone mineralization, and calcium absorption in wide range of bone disorders along with wide range of adverse health conditions, comprising cancer and certain autoimmune diseases.

Transportation Course of Macromolecules to the Nucleus from the Extracellular Environment: Steroid Hormones’ Cellular Entry Mode Revisited

Abstract: Polyomavirus virions such as simian virus 40 (SV40), antinuclear antibodies such as immunoglobulin G (IgG) and steroid hormones all enter the nucleus from the extracellular environment. Testosterone-bovine serum albumin conjugate labeled with 2 nm colloidal gold (testosterone-BSA-gold) is taken up by endocytosis into target cells, and enter the nucleus through a similar route as SV40 nuclear migration. Upon injection into the vascular system of rats, IgG coupled with hydrocortisone also enters the hormone-target cell nuclei with intact antigenicity. These results suggest that steroid hormones could act as transporters to deliver exogenous macromolecules, e.g. drugs, into their target cell nuclei in vivo, although further studies are required on whether steroid hormones coupled with proteins exert genomic actions in the nucleus, etc. Finally, testosterone-BSA-gold seems to be isolated from the cytosol in the processes of nuclear entry. Together, these findings challenge the popular belief that steroid hormones mostly enter the cell in unbound form via uncontrolled passive diffusion.

Sirtuin Activator and Inhibitor Affect Early Dictyostelium Development upon Starvation

Abstract: Sirtuin is an evolutionally conserved histone deacetylase. The effects of sirtuin activator, resveratrol and inhibitor, nicotinamide on early Dictyostelium development upon starvation were examined. The cell streaming was appeared at 7 h and tight aggregates was formed at 24 h in control cells under submerged culture. Resveratrol treatment accelerated the development. Cell streaming at 6 h and weak aggregates at 9 h were already observed. While, nicotinamide treatment delayed the development. The initiation of streaming was delayed at 9 h, and the development was still under a cell streaming even after 24 h. Dictyostelium development starts inducing the essential molecules adenylyl cyclase, aca and cAMP pulsing induces cell-surface cAMP receptor (carA). Resveratrol treatment significantly increased the expression level of aca at 4 and 6 h after starvation. Nicotinamide treatment decreased the aca expression level at 4 h and carA expression level at 6 h. The induction of aca upon starvation starts with the transcriptional activation by MybB, a. putative transcription factor. MybB expression recovered the initiation timing of the cell streaming against the effect of nicotinamide, but the aggregates formation was still impaired at 24 h. Resveratrol had no effects on MybB-null mutant cells. Neither streaming nor aggregates formation was observed even after 24 h with low levels of aca expression after starvation by resveratrol treatment. These findings suggest that sirtuins may participate in more than one point of the early developmental processes of Dictyostelium upon starvation.
